Lymphocyte subsets in healthy Saudi donors دراسة الخلايا الليمفاوية المختلفة في متبرعين أصحاء سعوديين

Abstract

إن الخلايا الليمفاوية تظهر اختلافات مع الأجناس المختلفة والسن وهدف هذا العمل هو دراسة الخلايا الليمفاوية المختلفة في السعودية (مكة المكرمة) ومقارنتها بالأجناس المختلفة وبدراسات سعودية أخري وهذه الدراسة قد تمت علي 54 شخص طبيعي وهم 13 رجل و45 سيدة بنسبة 1 إلي 3 وكانت أعمارهم تتراوح من 17 إلي 45 سنة ومتوسط أعمارهم هو 22 ± 58 سنة وتمت دراسة الخلايا الليمفاوية وأنواعها المختلفة بواسطة جهاز التدفق الخلوي بي دي وتم حساب العد المطلق للخلايا وأظهرت قيمة كل من السي دي 3 4 816 69 كنسبة مئوية وعد مطلق وتم مقارنتها بدراسات أخري (سعودية، إثيوبيا، ألمانيا، سنغافورة، هونج كونج)وأظهرت النتائج أن كل العد المطلق لسي دي المختلفة ذو دلالة إحصائية ذات قيمة عالية عندما قورنت بالدراسات الاخري فيما عدا السي دي 4 عند مقارنته بالدراسة الألمانية وأظهرت نسبة السي دي 4 إلي 8 ارتفاع ذو دلالة إحصائية عالية عندما قورنت بالدراسات الاخري فيما عدا دراسة سعودية سابقة التي أظهرت عدم اختلاف وانخفاض عند المقارنة بالدراسة الألمانية وعند مقارنة السي دي المختلفة في الرجال والنساء لم تظهر أي اختلافات ذات دلالة إحصائية فيما عدا زيادة نسبة السي دي 3 في النساء وزيادة العد المطلق لسي دي 69 في الرجال عن النساء ونستنتج من هذه الدراسة ان معظم العد المطلق لسي دي المختلفة (3 4 8 16 69) ونسبة السي دي 4 إلي 8 أظهرت ارتفاع ذو دلالة إحصائية مرتفعة عند مقارنتها بالدراسات الاخري ونقترح أن هناك عوامل وراثية وراء ظهور هذا الاختلاف وإنه يجب علي كل مجتمع أن يضع حدود طبيعية له وأيضا يجب أن تؤخذ عينات من أماكن مختلفة في المملكة العربية السعودية لتأكيد هذه النتائج

Background Lymphocyte and their subsets show variation with race and age Aim of the work The aim of this study was to detect the distribution of lymphocyte subsets in Saudi population (Makkah city) and to compare with different races and with previous Saudi studies Subjects and Methods This study was conducted on 54 healthy subjects ; they were 13 males and 41 females with male to female ratio of 1 3 Their ages ranged from 17 to 45 years with a mean age of (22 ± 58) years Lymphocyte and their subsets were analyzed by flow cytometry BD FACS Calibure and their absolute counts were calculated Results The CD3 CD4 CD8 CD16 and CD69 were expressed as percentage and absolute counts They were compared to other studies (Ethiopia Dutch Singapore Hong Kong and two previous Saudi studies) For all the studied CD markers all the absolute counts of the different CD markers had higher significant values (P < 001) as compared to other studies expect for CD4 of the Dutch study and CD8 of one Saudi study which showed no significant differences from our study (P > 005) The CD4 CD8 ratio had higher significant values when compared to other studies (P < 001) except for the Dutch study and another Saudi study which showed significant decrease (P < 005) and no significant difference (P > 005) when compared to our results respectively There were no significant differences when comparing the males to the females groups as regards percentage and absolute counts of the different CD markers studied except for significant increase of CD3 percentage in the females group (P < 005) and higher significant values of CD69 absolute counts in the males group (P < 001) Conclusion Most of the absolute counts of the different CD markers studied in our work (CD3 CD4 CD8 CD16 and CD69) and the CD4 CD8 ratio are higher when compared to different populations Our study suggests that there may be true genetic differences in lymphocyte subsets Every population must establish a normal range of their own ; it is recommended that samples from several other areas in Saudi Arabia should therefore be studied to confirm our data
